3. The sterilization of the culture material is not complete and it is contaminated by miscellaneous bacteria. The main reasons for incomplete sterilization are: 1) Improper sterilization operation. The sterilization method of normal pressure 100 ℃, 6-8 hours or high pressure 121 ℃, 0.5-2 hours can kill the mycelium and spores of general miscellaneous bacteria. At present, most of the edible fungus culture material adopts atmospheric pressure sterilization or high pressure sterilization method. Atmospheric pressure sterilization is more difficult to sterilize grains, dung grass and other materials, and it is not easy to achieve the effect of complete sterilization; while the effect of high pressure sterilization is better. 2) The culture material is different. Factors such as the type of materials required to make bacteria, the bacterial content, water content and pH of the culture material will affect the sterilization effect. The corresponding sterilization time should be selected according to the characteristics of different culture materials. 3) The influence of the bacteria packaging container. When making seeds, generally use bottles or bags to package the bacteria. Plastic bags transfer heat faster, while glass bottles transfer heat slower. When using the same culture material and the same sterilization method, the bottled material takes longer to sterilize than the bagged material.
4. The strains contain miscellaneous bacteria. The original species of edible fungus strains used for expanded culture must be pure bacteria, so as to ensure the singleness of the strains. If the edible fungus contains miscellaneous bacteria, it will lead to a decrease in the inoculation rate.
5. Improper operation of vaccination personnel. The inoculation process of edible fungi requires a strict aseptic operation environment. If the operation is not standardized, it is easy to cause the culture material to be contaminated by miscellaneous bacteria.
